Histidinová kináza 33 (Hik33) patří mezi nejlépe prostudované proteiny dvousložkového systému u sinic. Její hlavní funkcí je ochrana sinice, před stresem, pomocí regulace genové exprese. V minulosti byly vytvořeny a analyzovány mutantní kmeny sinice Synechocystis postrádající Hik33. Výsledky analýz se u jednotlivých publikací lišily, což je vzhledem k použití stejného kmenu zvláštní. Pravděpodobná příčina odlišných výsledků spočívá v tvorbě takzvaných revertantních kmenů, u kterých došlo vlivem stresu k tvorbě supresorovým mutacím. Diplomová práce je zaměřena na popis fenotypových vlastností mutanta Hik33, tvorbě a analýze revertantních kmenů a jejich supresorových mutací, a nakonec k objasnění struktury a vazebných jednotek histidinové kinázy 33 pomocí tvorby genového konstruktu hik33-Strep.

Histidine kinase 33 (Hik33) is one of the most well studied proteins of the two-component system in cyanobacteria. Its main function is to protect the cyanobacterium from stress by regulating gene expression. In the past, mutant strains of Synechocystis lacking Hik33 have been generated and analysed. The results of the analyses varied from publication to publication, which is strange given the use of the same strain. The probable cause of the different results lies in the formation of so-called revertant strains, in which suppressor mutations were generated due to stress. This thesis is aimed at describing the phenotypic properties of the Hik33 mutant, the formation and analysis of revertant strains and their suppressor mutations, and finally the elucidation of the structure and binding units of histidine kinase 33 by means of the formation of the hik33-Strep gene construct.
