This thesis develops and implements novel techniques for tracking nitrate isotopes to identify the major processes, including nitrification and denitrification, in the Bohemian National Forest's pristine Plešné, Čertovo, and Černé Lakes settings. The study optimizes a method for precise isotopic monitoring by conducting a comparative examination of three independent nitrous oxide conversion protocols. The selected method was applied to analyze the lake samples, providing crucial insights into the relative inputs and complicated nitrogen cycling mechanisms inside these unique forest lake ecosystems.

The head of the commission welcome the student and the members of the commission, all members of the commission except the supervisor were present. The student presented the nitrogen cycle, the accumulation of nitrogen in the biosphere, the studied protocols, sampling sites, the aims, methods used and results on the identified genes.
The supervisor and the opponent presented their reviews, and the student answered the opponent’s questions. The student then answered questions from the commission on the presence of TiCl4 in the TiCl3 preparation, on the 15N concentration in the atmosphere, and the d15N differences in the nitrification and denitrification processes.
