Notch signální dráha hraje důležitou roli při buněčné diferenciaci mnohobuněčných organismů a CSL protein je klíčovou složkou této dráhy. Tato práce je zaměřena na CSL protein Sphaerospora molnari, parazitického žahavce ze skupiny Myxozoa. Cíli byly optimalizace produkce rekombinantního CSL proteinu v expresním systému Escherichia coli, purifikace tohoto proteinu a produkce specifických protilátek proti purifikovanému CSL. Všech těchto cílů bylo dosaženo a práce byla zakončena úspěšným ověřením specifity protilátek proti purifikovanému CSL proteinu. Získaný rCSL a protilátky mohou být užity pro budoucí studie struktury CSL proteinu a jeho interakcí s DNA a imunoznačení, což může pomoci porozumět tomu, jak Notch dráha probíhá nejen u myxozoí, ale také u dalších časně se větvících metazoí.

Notch signaling pathway plays an important role in cell differentiation in multicellular organisms and the CSL protein is a key component of this pathway. This thesis focused on the CSL protein from a myxozoan parasite of the phylum Cnidaria Sphaerospora molnari. The objectives were to optimize the production of the recombinant protein in the Escherichia coli expression system, to purify this protein, and to produce specific antibody against the purified CSL. All these goals were achieved and the work ended with successful demonstration of the antibodies against the purified CSL protein. The rCSL and antibodies obtained will be used for future studies on the structure of the CSL protein, immunolabelling and its interactions with DNA, which may help to understand how the Notch pathway works not only in Myxozoa, but also in other early-branching metazoans.
