Course content
Content of lectures: 1. Absorption and emission spectroscopy, measurements of dispersive and spectral properties of monochromator. 2. Measurements of energy transfer between molecules by femtosecond laser spectroscopy. 3. Building acoustics, measurements of reverberation time and room acoustical parameters. 4. Experimental study of vacuum systems - measurement of pumping speed, leak detection 5. Magnetron sputtering deposition of thin films 6. Atomic force microscopy: high resolution imaging of surface topography in contact and intermittent contact mode, measurements of binding interactions between pairs of single molecules using dynamic force spectroscopy. 7. Single molecule spectroscopy: imaging of single molecules by a smFRET pair Content of practicals: Experiments and data analysis

Learning activities and teaching methods
Monologic (reading, lecture, briefing), Dialogic (discussion, interview, brainstorming), Laboratory
  • Class attendance - 50 hours per semester
  • Preparation for classes - 30 hours per semester
Learning outcomes
The course offers practical special measurements in physics, which goes beyond the techniques taught in practical courses of basic physics course. Lectures provide necessary theoretical and experimental basis. Experiments are carried out in research laboratories at the Faculty of Science.
The students will gain practical knowledge of selected advanced experimental methods commonly used in physics/biophysics. They will be able to design and carry out experiments using the scientific instruments used in the course.
Prerequisites
Passing all practical courses of general physics (Practicals 1-4)
UFY/CV013
----- or -----
UFY/FYZ3 and UFY/FPR3

Assessment methods and criteria
Student performance assessment, Analysis of student's work activities (technical works)

Carrying out all experimental work during the practicals and handing out protocols to all tasks.
