Course content
Content of lectures: The main topics of the course by individual weeks (13 weeks per semester) of teaching: New trends and challenges in plant biotechnology (1-2) Theoretical background of plant biotechnology (3-4) Biotechnology in plant breeding - in vitro techniques, genetic and phenotypic stability of in vitro cultures (5) Micropropagation and production of virus-free plants (6) Long term preservation of germplasm (7) Embyocultures in interspecific crossing (8) Haploid cultures and production of DH plants (9) Callus and suspension cultures, single cell cultivation, protoplast cultures (10) Recombinant DNA technology in plant biotechnology (11-13) Content of tutorials/seminar: Preparation of culture media, preparation and sterilization of plant material Establishment of callus culture from leaves Isolation of apical meristems Isolation of anthers and microspores Disc method of transformation, hypocotyl segments from in vitro seedlings, cocultivation with bacteria, transfer to culture medium and induction of callus culture

Learning outcomes
The aim of the course is to introduce the issue of plant cell, tissue and organ cultures in vitro, which represent the methodological basis of plant biotechnology and have applications in genetics and plant breeding, in the production of biotech (GMO) crops.

Credit: attendance at the laboratory practicals (max. 3 absences), completing laboratory protocols, presentation of seminar project - application of plant biotechnology. Written exam (min. 50 %)
