Course content
Lectures 1. The subject of analytical chemistry, sampling procedures, sample decomposition, preparation of samples for the analysis. 2. Qualitative analytical chemistry, sensitivity of analytical reactions, grouping of inorganic ions into classes, detection of selected ions. 3. Protolytic equilibria, pH, theory of acids and bases. 4. Gravimetric analyses, principles of titrations. 5. Volumetric analysis I (acidimetry, alkalimetry). 6. Volumetric analysis II (methods based on redox processes, precipitation titrations, complexometry). Classical chemical separation methods - distillation, extraction, ion-exchangers. 7. Differences between classical and instrumental analysis, potentiometry, pH measurements, ion-selective electrodes, conductometry. 8. The use of electrolytic processes in analytical chemistry, polarography, modern polarographic techniques. 9. Spectral and optical methods, emission optical analytical methods, fluorimetry. 10. Spectral and optical methods, atomic and molecular absorption spectrophotometry. 11. Infrared spectrometry and mass spectrometry. 12. Refractometry, polarimetry. Chemical separation methods, gas chromatography. 13. Chemical separation methods, liquid chromatography. 14. Electromigration methods, electrophoresis, isotachophoresis. Evaluation of analytical data. Practical exercises 1. Principles of safe work, common operations in chemical laboratory, toxicology, calculations 2. Qualitative analysis - detection of cations by the hydrogen sulfide procedure 3. Gravimetric determination of iron with the use of ammonium hydroxide 4. Acidimetry - preparation of a HCl titration solution and determination of the titre with the help of KHCO3, determination of the total alkalinity of technical grade hydroxide 5. Complexometric determination of calcium, magnesium and water hardness 6. Argentometric determination of iodides by Volhard 7. Bromatometric and iodometric determination of L-ascorbic acid 8. Manganometry - preparation of a KMnO4 volumetric solution and determination of the titre with the help of Mohr salt, determination of FeSO4 contents in the medicinal product Ferronat retard 9. Potentiometry - determination of chlorides in mineral water, measurement of pH 10. Spectrophotometry - preparation of calibration solutions, plotting of calibration curves, determination of Fe3+ with rhodanide

Learning outcomes
The target of the subject is to acquaint students with basic methods of classical and instrumental analysis. The lectures will provide students with an outline of analytical methods suitable for inorganic and organic chemical analysis of samples in the field of chemistry of environment, industry and biology. The target of practical exercises is to master basic skill in the work in laboratories of analytical chemistry.
The students will be capable of proper orientation in basic analytical techniques, will know principal possibilities of these methods and will be able to estimate the suitability of a relevant method for a given purpose. They will be able to communicate in qualified manner with specialists in the field of analytical chemistry.

Standard knowledge in chemistry at a level of secondary school oriented at general education or natural sciences

Completing laboratory exercises, compilation of protocols, mastering the scope of the study designed for the examination.
