Content of practicals: 1) Isolation of total genomic DNA, preparation of restriction genomic library 2) Preparation of SSR-enriched genomic library via hybridization with biotin-labelled oligonucleotides and capture by streptavidin coated magnetic beads 3) Cloning, amplification and sequencing of SSR-enriched genomic library 4) Design of SSR primers (based on sequences containing SSR loci)

Learning outcomes
|
Nuclear microsatellites (SSRs) are powerful and reliable tools for studies of genetic diversity. The aim of the course is to introduce students in development of specific SSR primers. The course is performed as a laboratory parctice. Students are encouraged to bring samples of their interest.

Prerequisites
|
basic understanding of scientific texts in English, basic knowledge of laboratory rules and molecular methods (PCR, DNA restriction and purification)

|
Development of laboratory protocols
understanding basics of the methods (ability to explain the technical principle), attendance at the practical part, completing laboratory protocols.
