Course content
|
Historical overview of the methodology plant tissue cultures. Aseptic cultures of plants, embryos, organs, tissues, cells and protoplasts. Principles of aseptic handling of plant material. Nutrition of in vitro cultures. Hormonal requirements of cultures. Physical cultivation conditions. Regulation of morphogenesis. Basic processes of regeneration in plant in vitro cultures. Genetic and phenotypic variability in in vitro cultures. Somaclonal variability. Production and use of haploid cultures and regenerants. Protoplast culture, somatic hybridization. Cryopreservation. Possibilities and prospects of application of biotechnology in crop improvement (cereals, pulses, oilseeds, potatoes, seed and root crops, legumes and forage grasses, vegetables, hops, ornamental plants, forest and fruit trees). Princeiples of DNA recombinant technology and plant genetic engineering. Transgenosis of plants via Agrobacterium. Integration of T-DNA into plant genome. T-DNA genes, the mechanism of T-DNA integration. Direct introduction of genes into plant genome. Transformation of protoplasts, tissue and organ culture, electroporation, microinjection, mikroprojectiles. Transgenes important for plant breeding. Ethical and legal issues of genetic engineering.
