Course content
Content of lectures: 1. Introduction to biochemical and molecular-biological studies (equipment and safety in the laboratory, GLP, experiment design, nucleic acids: structure and function, genes and genome, isolation and work with NA, PCR, sequencing, basics of bioinformatics) 2. Methods and use of recombinant DNA in biology (cloning of genes and cloning vectors, gene analysis and expression, expression of foreign genes and preparation of recombinant proteins, types of expression systems basics of cell cultures) 3. Proteins (protein function and structure, posttranslational modification, (enzymes: active sites and catalytic mechanism, control of enzymatic activity, utilization of enzymes), protein engineering, proteomics) 4. Electrophoretic methods (principles, media, agarose Electrophoresis (DNA, RNA and capillary electrophoresis) SDS-PAGE, native gels, gradient gels, 2D gels, zymography, Western blotting, immunobiochemistry) 5. Chromatography (Principles, Types of Chromatography, HPLC, Ion, Affinity, SEC, Adsorption, Distributing, Gas) 6. Centrifugation (principle and types of centrifuges, rotors, sedimentation principles, preparative centrifugation, analytical centrifugation) 7. Microscopy (introduction and types of microscopes, composition of microscopes, optical microscopy (techniques and limitations, resolution, sample preparation, stereomicroscope, confocal microscopy, fluorescence microscopy). Electron microscopy (types and techniques, preparation of samples, limitations), visualization in living cells and tissues 8. Mass spectrometry techniques 9. Spectroscopic methods in biochemistry Content of practicals: During the course the knowledge gained during the lectures will be enhanced.

Learning outcomes
The lectures provide students with biochemical methods to monitor cell life, and thus the function of countless proteins and other macromolecules, before structurally describing very complex molecules of enzymes, receptors, transporters, regulators, nucleic acids.
Student should understanding basic principles on techniques in molecular biology a biochemistry according to the specific lectures in the section "Obsah, opory".
Prerequisites
Basic knowledge in biology, biochemistry and biology.

Assessment methods and criteria
Student performance assessment, Test

Active participation on lectures, understanding of the subject. The progress is continuously checked during the course by group discussions. It is required min. 60 % in the final exam.
