Obsah předmětu
|
1) Overview and application of cell and molecular biology not only in aquaculture 2) Summary of the latest knowledge of fish reproduction biology 3) Use of germ stem cells in aquaculture (cryopreservation, in vitro cultivation and reproduction by surrogate parents) 4) Nuclear transfer (cloning) 5) Procedures and utilization of gene and genomic manipulations (polyploidy, gynogenesis, androgenesis, gene transfer and editing); Possibilities of induction and exploitation of gender reversal and production of monosexuals 6) Biochemical and molecular methods (genomics, transcriptomics and proteomics) 7) Basics of population genetics (genetic variability, molecular genetic markers in population genetics, Hardy-Weinberg equilibrium, individuals vs. aquaculture animals) 8) Gene-mapping, QTL, marker-assisted selection. Training will be based on topics of lectures. Students will practice micromanipulation (embryo injection, labeling, isolation and transplantation of germ stem cells), basic molecular techniques (extraction of whole genome DNA from tissues, PCR amplification of selected fragments, PCR modification, fragment analysis, sequencing etc. ) and basic molecular genetic data processing and population analysis programs.

The aim of this course is to acquaint students with the use of modern cellular and molecular approaches in aquaculture.
Student will get basic knowledge related to current biotechnological applications in aquaculture. Student will theoretically manage biochemical and molecular methods in aquaculture, genomic manipulations, cloning, chimerism induction, sex reversals, fundamentals of population and conservation genetics and some of them will try to practise.

Requirement for credit: Students are required to pass at least 80% of final test, they have 3 attempts. Requirement for exam: oral examination.
